Controlled release manufacturing

Catalent Pharma Solutions has a range of experience with innovative and traditional controlled release technologies:

EnCirc® pellets for higher drug loading:

EnCirc® pellets can be used for immediate and controlled-release capsules or tablets.

Attributes
High-potency pellets
Predictable mean-size and narrow-size distribution
High sphericity
Low friability
Benefits
Typical drug loading exceeds 80%
Reproducible drug-release profiles
Uniformity of dose
Reproducible drug-release profiles
Uniform coatings due to fewer surface imperfections than nonpareils
Durable product that withstands rigorous coating processes

EnVel® system for taste masking:

EnVel® is a system of proprietary taste-masking technologies for chewable tablets. The active ingredient is enveloped as a matrix using pharmaceutically acceptable excipients or the active is isolated as discrete particles using a hydrocolloid polymer.

EnVel® technologies can greatly improve patient acceptance of Rx or OTC chewable tablets. The drug-dissolution profile remains unchanged.

Attributes
Effective taste masking
Unique drug-polymer matrix
Higher drug loading
Drug-particle isolation barriers
Benefits
Greatly improved palatability of a wide variety of active ingredients
Taste-masking properties are not affected by chewing or compression
Smaller dosage forms
Improved patient compliance
Drug particles are isolated from fluids during chewing

EnSolv® for improved dissolution:

EnSolv® technologies can provide a formulation solution for new chemical entities as well as extend life cycles of many marketed products.

Two EnSolv® technologies are available to modify certain drug crystals by way of:

  • solid dispersion using a hot-melt process, or
  • aqueous deposition

EnSolvĀ® for solid dispersion:

A hot-melt technology that transforms the drug crystals into a higher-energy amorphous state, thus providing improved dissolution of an insoluble active ingredient.

Attributes
Improved dissolution
Unlocks the potential of poorly soluble drugs by allowing a stable supersaturated solution to form
Benefits
Fine-particle solid ?dispersion
Stable, fine powder for compression or encapsulation

EnSolv® for aqueous deposition:

A patented co-processing technology that generates a suspension in the stomach, and is for use in immediate-release capsules or tablets.

Attributes
Faster dissolution
Performs similar to a suspension
Improved content uniformity
Improved stability by coating
Improved compressibility
Higher drug loading
Improved material flow
Benefits
Unlocks the potential of poorly soluble drugs
Rapid and complete exposure of high drug-surface area
Consistently meets specifications
Withstands further processing and severe storage conditions
Improves processing for difficult-to-formulate actives
Increases dosing alternatives
Improves processing for difficult-to-formulate actives

Particle coatings for controlled release:

Custom modified-release formulations are specifically designed to meet the desired drug release profile.

Benefits:
Incompatibility barrier
Taste masking
Delayed or controlled release
Enteric coating
Post gastric release
