Title: Breaking the Oral Delivery Barrier for Macromolecules: From Dissolution to Delivery

Oral delivery of macromolecules remains highly challenging due to acidic degradation in the gastrointestinal tract, enzymatic shredding and limited permeability. Recent innovations in delayedrelease softgels are expanding the formulation toolbox to address these barriers. In particular, delayedrelease softgels protect against gastric degradation and enable controlled release in specific intestinal regions. This approach allows release kinetics to align with transient permeability enhancement, thereby enhancing bioavailability while preserving patient-preferred oral dosing. This presentation will examine key scientific and biopharmaceutical barriers to oral macromolecule delivery and highlight dissolution-relevant learnings from delayed-release softgel technologies, using select recent Catalent internal data.
